Low-binding plasticware reduces loss of dilute solutions, and sterile filtration is used when a preparation must remain free of microbial growth. Records of batch number, reconstitution date and storage history are what allow a later analytical result to be interpreted meaningfully.

In humans, fatty acids are formed from carbohydrates predominantly in the liver and adipose tissue, as well as in the mammary glands during lactation. The pyruvate produced by glycolysis is an important intermediary in the conversion of carbohydrates into fatty acids and cholesterol. This occurs via the conversion of pyruvate into acetyl-CoA in the mitochondrion. However, this acetyl-CoA needs to be transported into cytosol where the synthesis of fatty acids and cholesterol occurs. This cannot occur directly. To obtain cytosolic acetyl-CoA, citrate (produced by the condensation of acetyl-CoA with oxaloacetate) is removed from the citric acid cycle and carried across the inner mitochondrial membrane into the cytosol. There it is cleaved by ATP citrate lyase into acetyl-CoA and oxaloacetate. The oxaloacetate is returned to mitochondrion as malate (and then converted back into oxaloacetate to transfer more acetyl-CoA out of the mitochondrion). The cytosolic acetyl-CoA is carboxylated by acetyl-CoA carboxylase into malonyl-CoA, the first committed step in the synthesis of fatty acids.

=== Stem cells === Levels of p53 play an important role in the maintenance of stem cells throughout development and the rest of human life. In human embryonic stem cells (hESCs)s, p53 is maintained at low inactive levels. This is because activation of p53 leads to rapid differentiation of hESCs. Studies have shown that knocking out p53 delays differentiation and that adding p53 causes spontaneous differentiation, showing how p53 promotes differentiation of hESCs and plays a key role in cell cycle as a differentiation regulator. When p53 becomes stabilized and activated in hESCs, it increases p21 to establish a longer G1. This typically leads to abolition of S-phase entry, which stops the cell cycle in G1, leading to differentiation. Work in mouse embryonic stem cells has recently shown however that the expression of P53 does not necessarily lead to differentiation. p53 also activates miR-34a and miR-145, which then repress the hESCs pluripotency factors, further instigating differentiation. In adult stem cells, p53 regulation is important for maintenance of stemness in adult stem cell niches. Mechanical signals such as hypoxia affect levels of p53 in these niche cells through the hypoxia inducible factors, HIF-1α and HIF-2α. While HIF-1α stabilizes p53, HIF-2α suppresses it. Suppression of p53 plays important roles in cancer stem cell phenotype, induced pluripotent stem cells and other stem cell roles and behaviors, such as blastema formation. Cells with decreased levels of p53 have been shown to reprogram into stem cells with a much greater efficiency than normal cells.
This increase is comparable to the increase which was found in mouse myotubes. In contrast to the mouse myotubes calciseptine did not change the reversal potential of the Ca2+ current. In rat muscle fibers after applying calciseptine there were very slight changes found in twitch tension, which shows us that the peptide had very little effect on muscle contraction. Calciseptine also had little effect on frog muscle fibers. There is a small effect of calciseptine during a series of repetitive stimuli which generate a tetanus; calciseptine increased tetanic tension. The average tetanic tension after applying calciseptine was slightly higher than the control value. Though calciseptine gives an increase in charge movement and increase in influx of Ca2+ through L-type channels this has no major effects on contraction of the muscle, even in tetanus. Research on rats and guinea pigs revealed that synthetic calciseptine and FS2 as well, have the same effect as their natural counterparts.

A liquid coolant—typically either liquid nitrogen (−196 °C, 77ºK, −320 °F) or 99% ethanol chilled with dry ice (−72 °C, 201ºK, −98 °F)—is used to bring the brand head down to its working temperature in an appropriately insulating container. Other coolants historically used with dry ice have included acetone, isopropanol, methanol, gasoline, kerosene and jet fuel. There is some anecdotal evidence that the lower viscosity of chilled acetone makes for whiter brands, presumably through better heat conduction. Experimental work has also been carried out using the direct application of coolants to an animal's skin. Freons 12, 21, 22 and 31 have all been evaluated as direct coolants, either as a spray or a slushy mixture of freon ice and liquid. Freons were chosen because they can be bought in pressurized cans that produce cryogenic temperatures when vented. This convenience was desirable aboard a ship or amid pinniped colonies to tag sea mammals for scientific study. Direct freezing with CFCs proved cumbersome and difficult to control, requiring custom masks to form an intentional pattern. The method has largely been abandoned since the Vienna Convention. Cryogenic alcohol baths readily absorb water from atmospheric humidity, producing slush. This slush is a poor conductor of heat and will ruin brands. If alcohol baths are to be used for a series of brandings the alcohol must be replaced every two hours from a fresh 99% supply. This hygroscopic phenomenon was the original impetus to seek non-water soluble coolants such as kerosene and jet fuel during the technique's development.

Xylazine is a potent α2-adrenergic receptor agonist. When xylazine and other α2-adrenergic receptor agonists are administered, they distribute throughout the body within 30 to 40 minutes. Due to xylazine's highly lipophilic nature, it directly stimulates central α2-adrenergic receptors as well as peripheral α-adrenergic receptors in a variety of tissues. As an agonist, xylazine reduces release of norepinephrine in the central nervous system. It does so by mimicking norepinephrine in binding to the pre-synaptic surface autoreceptors, which leads to feedback inhibition of norepinephrine release. Recent data suggests that xylazine treatment can induce dopamine release in the nucleus accumbens through an unresolved mechanism, and this effect is blocked by atipamezole. Xylazine also serves as a transport inhibitor by suppressing norepinephrine transport function through competitive inhibition of substrate transport. Accordingly, xylazine significantly increases Km and does not affect Vmax. This likely occurs by direct interaction on an area that overlaps with the antidepressant binding site. For example, xylazine and clonidine suppress uptake of iobenguane (MIBG), a norepinephrine analogue, in neuroblastoma cells. Xylazine's chemical structure closely resembles clonidine. It has also been reported that xylazine activates the κ-opioid receptors, with low potency, which may contribute to its effects. Unlike other α2-adrenergic receptor agonists xylazine does not have any imidazoline receptor activity.
