== Formation == Initial funding for the formation of Onyx came from biotechnology firm Chiron Corporation (granted a 43% stake in the new company) and venture capital investors: Avalon Ventures, Institutional Venture Partners (IVP), J. H. Whitney & Company and Kleiner Perkins. == Pharmacology == The drug is a selective KOR antagonist and shows approximately 10- to 20-fold higher affinity for the KOR (Ki = 1–3 nM) over the μ-opioid receptor (MOR) (Ki = 10–64 nM) and has negligible affinity for the δ-opioid receptor (DOR) (Ki > 4,000 nM). It is a "short-acting" or non-inactivating antagonist of the KOR (as opposed to irreversible antagonists like JDTic). Although originally characterized as a KOR neutral antagonist however, subsequent research revealed in 2020 that PF-04455242 is actually only a moderately efficacious partial antagonist of the KOR (Imax ≈ 50%). In any case, the drug reversed the analgesic and prolactin-elevating effects of the KOR agonist spiradoline in animals, showed efficacy in animal models predictive of antidepressant activity, and reversed stress-induced reinstatement of cocaine-seeking behavior. However, PF-04455242 also showed a variety of other weak off-target activities.

The female is gravid for 50 to 59 days. The king cobra is the only snake that builds a nest using dry leaf litter, starting from late March to late May. The parthenogenetic mechanism appears to be a variation of meiosis referred to as terminal fusion automixis in which there is fusion of the meiotic products formed at the anaphase II stage of meiosis. The venom of hatchlings is as potent as that of the adults. They may be brightly marked, but these colours often fade as they mature. They are alert and nervous, being highly aggressive if disturbed. The average lifespan of a wild king cobra is about 20 years.
During an episode of The Adam Carolla Podcast that aired on June 6, 2012, Marrow claimed that after being discharged from the Army, he began a career as a bank robber. ==== Inflammation ==== When particular GPCRs are activated by their specific chemokines Gβγ directly activates PI3Kγ which is involved in the recruitment of neutrophils that contribute to inflammation. It has been discovered that the inhibition of PI3Kγ significantly reduces inflammation. PI3Kγ is the intended target molecule in the prevention of inflammation as it is the common signalling effector of many different chemokine and receptor types involved in promoting inflammation. Although PI3Kγ is the intended target there are other isoforms of PI3 which perform different functions from PI3Kγ. Since PI3Kγ is specifically regulated by Gβγ, while other isoforms of PI3 are largely regulated by other molecules, inhibiting Gβγ signalling would provide the desired specificity of a therapeutic agent designed to treat inflammation.
