=== Body wall === The body wall layers include a thin cuticle covering, an epidermis consisting of a single layer of cells, a thick dermis formed of connective tissue, a thin coelomic myoepithelial layer for the muscles, and a peritoneum which lines the body cavity. The dermis contains an endoskeleton of calcium carbonate components known as ossicles. These are honeycomb-like structures composed of calcite microcrystals arranged in a lattice. They vary in form, from flat plates to granules to spines, and cover the aboral (top) surface. Some are specialised structures such as the madreporite (the entrance to the water vascular system), pedicellariae, and paxillae. Paxillae are umbrella-like structures found on starfish that live buried in substrate. == Clinical significance == Adult human blood normally contains three types of hemoglobin: hemoglobin A, which makes up approximately 95% of the total; hemoglobin A2, which accounts for less than 3.5%; and a minute amount of hemoglobin F. If abnormal hemoglobin variants such as hemoglobin S (which occurs in sickle cell disease), C or E are present, they will appear as unexpected bands on electrophoresis (provided they do not migrate to the same place as other hemoglobins). Hemoglobin electrophoresis can also be used to investigate thalassemias, which are caused by decreased production of subunits of the hemoglobin molecule. Hemoglobin A2 levels are typically elevated in beta-thalassemia minor and hemoglobin F may be slightly increased. In beta-thalassemia major, hemoglobin A is decreased (or in some cases absent) and hemoglobin F is markedly elevated; A2 levels are variable. In hemoglobin H disease, a form of alpha-thalassemia, an abnormal band of hemoglobin H can be detected, and sometimes a band of Hemoglobin Barts; but in the milder alpha-thalassemia trait, electrophoresis results are effectively normal.

== Laboratory methods == Hemoglobin A2 has a crucial role in screening and diagnosis of the beta-thalassemia trait. Various laboratory methods exist, each providing different outcomes of accuracy. The different quantification methods of HbA2 relies on how effective it can be separated from other hemoglobin variants. Various laboratory methods are employed, such as cation exchange high-performance liquid chromatography (HPLC), microcolumn chromatography, and cellulose acetate electrophoresis with elution. At first, cellulose acetate electrophoresis was utilized to measure HbA2, but this process proved to be too time-consuming and labor-intensive, making it impractical for large examinations or samples. Similar efficiency issues were encountered with other methods like IEF and scanning densitometry. These two methods separate proteins based on their isoelectric point. Chromatography, another commonly used method, demonstrated reliability in diagnosing individuals with the beta carrier gene. === Engineered peptidyl transferase center === In 2014, it was shown that by altering the peptidyl transferase center of the 23S rRNA, ribosomes could be created which draw on orthogonal pools of tRNA. The 3' end of tRNAs is universally conserved to be CCA. The two cytidines base pair with two guanines the 23S rRNA to bind the tRNA to the ribosome. This interaction is required for translational fidelity. However, by co-mutating the binding nucleotides in such a way, that they can still base pair, the translational fidelity can be conserved. The 3'-end of the tRNA is mutated from CCA to CGA, while two cytidine nucleotides in the ribosomes A- and P-sites are mutated to guanidine. This leads to ribosomes which do not accept naturally occurring tRNAs as substrates and to tRNAs, which cannot be used as substrate by natural ribosomes. To use such tRNAs effectively, they would have to be aminoacylated by specific, orthogonal aaRSs. Most naturally occurring aaRSs recognize the 3'-end of their corresponding tRNA. aaRSs for these 3'-mutated tRNAs are not available yet. Thus far, this system has only been shown to work in an in-vitro translation setting where the aminoacylation of the orthogonal tRNA was achieved using so called "flexizymes".
