== History == Daptomycin, originally designated as LY 146032, was discovered by researchers at Eli Lilly and Company in the late 1980s from the actinomycete Streptomyces roseosporus. LY 146032 showed promise in phase I/II clinical trials for treatment of infection caused by Gram-positive organisms. Lilly ceased development because high-dose therapy was associated with adverse effects on skeletal muscle, including myalgia. The rights to LY 146032 were acquired by Cubist Pharmaceuticals in 1997, which following U.S. Food and Drug Administration (FDA) approval in September 2003, for use in people older than 18 years, began marketing the drug under the trade name Cubicin. Cubicin is marketed in the EU and in several other countries by Novartis following its purchase of Chiron Corporation, the previous licensee. In July 2016 the FDA approved a reformulated version of daptomycin, designated Cubicin RF, which is stored at controlled room temperature rather than requiring refrigeration. === The nucleic acid template hypotheses === Alexander Dounce, of the University of Rochester Medical Center in New York, came close to the correct interpretation of protein synthesis from RNA. In 1952, he proposed the first notion, naming it the "nucleic acid template hypotheses," that proteins were made from RNA, rather than from DNA. However, since the exact chemical structure of DNA or RNA was not known at the time, he suggested erroneously that proteins and RNA have complimentary sequences, as he described in Enzymologia: "the specific arrangement of amino acid residues in a given peptide chain is derived from the specific arrangement of nucleotide residues in a corresponding specific nucleic acid molecule." It was met with criticism in Nature the next year, to which Dounce defended his hypothesis:In the hypothesis proposed by me, ribonucleic acid was deliberately chosen as the template, since it seems probable from a number of considerations and experimental investigations that protein syn- thesis may occur in cytoplasm as well as in nuclei. === Effects of Temperature === All enzymes work within a range of temperature specific to the organism. Increases in temperature generally lead to increases in reaction rates. There is a limit to the increase because higher temperatures lead to a sharp decrease in reaction rates. This is due to the denaturating (alteration) of protein structure resulting from the breakdown of the weak ionic and hydrogen bonding that stabilize the three-dimensional structure of the enzyme active site. The "optimum" temperature for human enzymes is usually between 35 and 40 °C. The average temperature for humans is 37 °C. Human enzymes start to denature quickly at temperatures above 40 °C. Enzymes from thermophilic archaea found in the hot springs are stable up to 100 °C. However, the idea of an "optimum" rate of an enzyme reaction is misleading, as the rate observed at any temperature is the product of two rates, the reaction rate and the denaturation rate. If you were to use an assay measuring activity for one second, it would give high activity at high temperatures, however if you were to use an assay measuring product formation over an hour, it would give you low activity at these temperatures.
== Notable incidents == In July 2009, Richard Torres, a correction officer at FMC Rochester, was indicted for smuggling contraband into the facility for an inmate in exchange for thousands of dollars in bribes. The contraband included cellular telephones, tobacco and creatine powder. Torres was terminated. Two months later, he pleaded guilty to soliciting a bribe and he was sentenced to one year in federal prison.
