He is Professor of Psychology in Psychiatry at the Perelman School of Medicine at the University of Pennsylvania, where he served as Director of the Center for Weight and Eating Disorders for over two decades. From 2011 to 2021, he held the endowed Albert J. Stunkard Professorship in Psychiatry. Wadden has published more than 500 peer-reviewed scientific papers and edited seven volumes on obesity and eating disorders. He has contributed to national health policy on obesity management, serving on expert panels for the National Institutes of Health, the Federal Trade Commission, the Department of Veterans Affairs, and the United States House of Representatives. He is a fellow of The Obesity Society. In 2015, The Obesity Society created the Thomas A. Wadden Award for Distinguished Mentorship, recognizing his education of scientists and practitioners in the field of obesity.
== Efficiency == The main limitation in the use of MLC is the reduction in efficiency (peak broadening) that is observed when purely aqueous micellar mobile phases are used. Several explanations for the poor efficiency have been theorized. Poor wetting of the stationary phase by the micellar aqueous mobile phase, slow mass transfer between the micelles and the stationary phase, and poor mass transfer within the stationary phase have all been postulated as possible causes. To enhance efficiency, the most common approaches have been the addition of small amounts of isopropyl alcohol and increase in temperature. === Pharmacodynamics === 3,4-DMA has been assessed in various biochemical and preclinical studies. Its affinity (Ki) for the rat serotonin 5-HT2A receptor has been assessed and was found to be 43,300 nM. For comparison, the affinity of para-methoxyamphetamine (PMA) was 33,600 nM, of 2,5-dimethoxyamphetamine (2,5-DMA) was 5,200 nM, and of 2,5-dimethoxy-4-methylamphetamine (DOM) was 100 nM in the same study. 3,4-DMA also showed affinity for the 5-HT1 receptor (Ki = 64,600 nM). The drug has additionally been found to be a monoamine oxidase inhibitor (MAOI), with an IC50Tooltip half-maximal inhibitory concentration of 20,000 nM for monoamine oxidase A (MAO-A), whereas it was inactive at monoamine oxidase B (MAO-B) (IC50 > 100,000 nM). 3,4-DMA does not produce hyperlocomotion and hence lacks stimulant-like effects in rodents. It also fails to produce stimulus generalization to dextroamphetamine in rodent drug discrimination tests, likewise suggesting that it lacks stimulant- or amphetamine-like effects. == Constitutionality in the United States == In 2006, the Supreme Court ruled in Hill v. McDonough that death-row inmates in the United States could challenge the constitutionality of states' lethal injection procedures through a federal civil rights lawsuit. Since then, numerous death-row inmates have brought such challenges in the lower courts, claiming that lethal injection as practiced violates the ban on "cruel and unusual punishment" found in the Eighth Amendment to the United States Constitution. Lower courts evaluating these challenges have reached opposing conclusions. For example, courts have found that lethal injection as practiced in California, Florida, and Tennessee is unconstitutional. Other courts have found that lethal injection as practiced in Missouri, Arizona, and Oklahoma is constitutionally acceptable. As of 2014, California has nearly 750 prisoners condemned to death by lethal injection despite the moratorium imposed when, in 2006 a federal court found California's lethal injection procedures to be unconstitutional. A newer lethal injection facility has been constructed at San Quentin State Prison which cost over $800,000, but it has yet to be used because a state court found that the California Department of Corrections and Rehabilitation violated the California Administrative Procedure Act by attempting to prevent public oversight when new injection procedures were being created. On September 25, 2007, the United States Supreme Court agreed to hear a lethal-injection challenge arising from Kentucky, Baze v. Rees.

== Mechanism of action == Normally, upon injury to the endothelium, collagen mediated GPVI signalling increases platelet formation by thromboxane A2, therefore creating a blood clot. In case of blood vessel damage, collagen on the extracellular matrix is exposed. As platelets interact with it, an activation signal is sent for aggregation. Platelets interact indirectly with collagen, via the von Willebrand Factor (vWF), which connects the collagen to the platelet GPIb receptor, forcing them close to the site of vessel damage. There, they can interact with receptors on the extracellular matrix, which stimulate adhesion through integrins (heterodimer α2β1), and downstream signalling. GPVI is present as a complex with the Fc receptor (FcR) γ-chain, which gets phosphorylated by SYK as a result of activation by a stimulus. This generates a downstream signal, leading to platelet activation. While this is important in case of injury, inappropriate activation of platelets can lead to the formation of clots within the circulation. Such is the case with Convulxin, which can induce a signalling cascade similar to that of collagen. Due to its high affinity, convulxin bind to GPVI and causes clustering of the glycoproteins. Research has proved that GPlb is not involved in convulxin-induced activation, but that the p62/GPVI collagen receptor is the unique binding site, and protein phosphorylation happens more rapidly and more intensely than in the case of collagen.

=== Validity === Donald Berry, writing in the journal Nature, has called attention to potential problems with the validity of ways in which many of the standardised tests are performed;[subscription required] in his article, as described in an accompanying editorial, Berry argues that anti-doping authorities have not adequately defined and publicized how they arrived at the criteria used to determine whether or not a test result is positive [which are] ...calibrated in part by testing a small number of volunteers taking the substance in question. [Berry argues] ...that individual labs need to verify these detection limits in larger groups that include known dopers and non-dopers under blinded conditions that mimic what happens during competition.
